ABA regulation of post-germination desiccation tolerance in wheat cultivars contrasting in drought tolerance Anais da ABC (AABC)
KOUR,SATINDER; ZHAWAR,VIKRAMJIT K..
ABSTRACT Post-germination desiccation tolerance (DT) was studied in two wheat cultivars. Effect of pretreatment of abscisic acid (ABA)/ osmotic/ salt/ heat stress was also studied. One day (d)-old seedlings of wheat cultivars PBW 644 (drought tolerant) and PBW 343 (drought susceptible) were exposed to ABA/stress treatment for next 1 d, desiccated for 4 d and subsequently rehydrated for 4 d. Biomass, protein, water, protein carbonyls (oxidative toxicity) and nitric oxide (NO) levels were measured in 2 d (treated), 6 d (desiccated), 10 d (rehydrated) seedlings. Vegetative reactive oxygen species (ROS)/ NO-pathways were studied under normal condition and ABA supply by supplying ROS/NO scavengers. Desiccation caused water loss and increased oxidative toxicity....

Can WRKY transcription factors help plants to overcome environmental challenges? Genet. Mol. Biol.
Finatto,Taciane; Viana,Vívian Ebeling; Woyann,Leomar Guilherme; Busanello,Carlos; Maia,Luciano Carlos da; Oliveira,Antonio Costa de.
Abstract WRKY transcription factors (TFs) are responsible for the regulation of genes responsive to many plant growth and developmental cues, as well as to biotic and abiotic stresses. The modulation of gene expression by WRKY proteins primarily occurs by DNA binding at specific cis-regulatory elements, the W-box elements, which are short sequences located in the promoter region of certain genes. In addition, their action can occur through interaction with other TFs and the cellular transcription machinery. The current genome sequences available reveal a relatively large number of WRKY genes, reaching hundreds of copies. Recently, functional genomics studies in model plants have enabled the identification of function and mechanism of action of several WRKY...

Changes in hormonal balance as key to reserve degradation after dormancy overcoming in Annona macroprophyllata and Annona purpurea seeds Rev. Bras. Frutic.
Ferreira,Gisela; De-la-Cruz-Chacón,Ivan; González-Esquinca,Alma Rosa.
Abstract Studies on the effect of the applying of phytoregulators to overcome seed dormancy have been carried out in Annonaceae species, thus the endogenous relationship between abscisic acid and gibberellins after the application of phytoregulators needs to be known in these species. Considering the importance of hormonal balance in dormancy mechanisms, the aim of this research was to elucidate how endogenous ABA and GA concentrations change after the application of phytoregulators in seeds and how these changes affect reserve degradation during germination of Annona macroprophyllata and A. purpurea seeds. Seeds were submitted to three conditions: - no soaking, soaking-in-water and soaking-in-GA4+7+benzyladenine. ABA, GA, lipids, proteins, and total...

Drying and reduction in sensitivity to desiccation of seeds of Alibertia edulis: the influence of fruit ripening stage Anais da ABC (AABC)
CREMON,THAIS; DRESCH,DAIANE M.; SCALON,SILVANA P.Q.; MASETTO,TATHIANA E..
ABSTRACT The intense environmental degradation in tropical regions suitable for agriculture has decreased native forest populations of plants with important fruits and medicinal properties. Alibertia edulis is a native tree from the Brazilian Cerrado. Knowledge about the effects of drying and storage on the physiological behavior of its seeds may aid in its sustainable exploitation and conservation. The goal of the present study was to determine which fruit ripening stage results in A. edulis seeds with higher tolerance to desiccation, and to investigate the effectiveness of polyethylene glycol (PEG) induced osmotic stress in combination with abscisic acid (ABA) in increasing seed desiccation tolerance during storage. Seeds were dried in activated silica...

Effect of abscisic acid on the mobilisation of galactomannan and embryo development of Sesbania virgata (Cav.) Pers. (Leguminosae - Faboideae) Rev. Bras. Bot.
POTOMATI,ADRIANA; BUCKERIDGE,MARCOS S..
Galactomannans (GM) are storage cell wall polysaccharides present in endospermic seeds of legumes. They are thought to be storage polymers, since it has been observed for a few species (among them Sesbania virgata) that they are completely broken down after germination and their products are transferred to the growing embryo. We examined the effect of 10-4 M abscisic acid (ABA) on the degradation of galactomannan in isolated endosperms and intact seeds of S. virgata. We found that after seed germination the initial embryo growth was retarded. Ultrastructural analysis showed that the embryo is completely surrounded by an endosperm which displays very thick galactomannan-containing cell walls. Although an inhibitory effect has been observed on the increase...

Ethylene and abscisic acid in the control of development of the rhizome of Kohleria eriantha (Benth.) Hanst. (Gesneriaceae) Braz. J. Plant Physiol.
Almeida,Julieta Andrea Silva de; Kascheres,Concetta; Pereira,Maria de Fátima D.A..
Kohleria eriantha has rhizomes which are underground stems with buds enclosed by modified leaves, that store starch. The buds of this rhizome can develop in two morphological patterns: an aerial shoot (similar to the mother plant) or a rhizome, depending on the water level present in the substrate. Development of the shoot was inhibited by low water availability (1 mL) in the substrate. It was verified that ethylene and ABA were involved in controlling the development of the rhizome pattern under low water availability. Treatments with ethrel, PEG and ABA induced shoot development whereas treatments with AgNO3 or AVG inhibited shoot development. Increased ethylene and ABA were observed under conditions that inhibited shoot development. Moreover, it is...

Eucalyptus ESTs involved in the production of 9-cis epoxycarotenoid dioxygenase, a regulatory enzyme of abscisic acid production Genet. Mol. Biol.
Guerrini,Iraê A.; Trigueiro,Rodrigo M.; Leite,Regina M.; Wilcken,Carlos F.; Velini,Edivaldo D.; Mori,Edson S.; Furtado,Edson L.; Marino,Celso L.; Maia,Ivan G..
Abscisic acid (ABA) regulates stress responses in plants, and genomic tools can help us to understand the mechanisms involved in that process. FAPESP, a Brazilian research foundation, in association with four private forestry companies, has established the FORESTs database (<A HREF="https://forests.esalq.usp.br">https://forests.esalq.usp.br</A>). A search was carried out in the Eucalyptus expressed sequence tag database to find ESTs involved with 9-cis epoxycarotenoid dioxygenase (NCED), the regulatory enzyme for ABA biosynthesis, using the basic local BLAST alignment tool. We found four clusters (EGEZLV2206B11.g, EGJMWD2252H08.g, EGBFRT3107F10.g, and EGEQFB1200H10.g), which represent similar sequences of the gene that produces NCED. Data...

Neospora caninum causes abortion and stillbirth in cattle. Identification of effective drugs against this parasite remains a challenge. Previous studies have suggested that disruption of abscisic acid (ABA)-mediated signaling in apicomplexan parasites such as Toxoplasma gondii offers a new drug target. In this study, the ABA inhibitor, fluridone (FLU), was evaluated for its action against N. caninum. Production of endogenous ABA within N. caninum was confirmed by ultra-performance liquid chromatography-tandem quadruple mass spectrometry. Subsequently, FLU treatment efficacy was assessed using in vitro. Results revealed that FLU inhibited the growth of N. caninum and T. gondii in vitro (IC50 143.1 +/- 43.96 mu M and 330.6 +/- 5238 mu M, respectively)....

Interactions between leaf water potential, stomatal conductance and abscisic acid content of orange trees submitted to drought stress Braz. J. Plant Physiol.
Gomes,Mara de Menezes de Assis; Lagôa,Ana Maria Magalhães Andrade; Medina,Camilo Lázaro; Machado,Eduardo Caruso; Machado,Marcos Antônio.
Thirty-month-old 'Pêra' orange trees grafted on 'Rangpur' lemon trees grown in 100 L pots were submitted to water stress by the suspension of irrigation. CO2 assimilation (A), transpiration (E) and stomatal conductance (g s) values declined from the seventh day of stress, although the leaf water potential at 6:00 a.m. (psipd) and at 2:00 p.m. (psi2) began to decline from the fifth day of water deficiency. The CO2 intercellular concentration (Ci) of water-stressed plants increased from the seventh day, reaching a maximum concentration on the day of most severe stress. The carboxylation efficiency, as revealed by the ratio A/Ci was low on this day and did not show the same values of non-stressed plants even after ten days of rewatering. After five days of...

Network analysis of ABA-dependent and ABA-independent drought responsive genes in Arabidopsis thaliana Genet. Mol. Biol.
Liu,Shiwei; Lv,Zongyou; Liu,Yihui; Li,Ling; Zhang,Lida.
Abstract Drought is one of the most severe abiotic factors restricting plant growth and yield. Numerous genes functioning in drought response are regulated by abscisic acid (ABA) dependent and independent pathways, but knowledge of interplay between the two pathways is still limited. Here, we integrated transcriptome sequencing and network analyses to explore interplays between ABA-dependent and ABA-independent pathways responding to drought stress in Arabidopsis thaliana. We identified 211 ABA-dependent differentially expressed genes (DEGs) and 1,118 ABA-independent DEGs under drought stress. Functional analysis showed that ABA-dependent DEGs were significantly enriched in expected biological processes in response to water deprivation and ABA stimulus,...

Physiological and cytological aspects of Inga vera subsp. affinis embryos during storage Braz. J. Plant Physiol.
Faria,José M.R.; Davide,Lisete C.; Silva,Edvaldo A. A. da; Davide,Antonio C.; Pereira,Roselaine C.; van Lammeren,André A.M.; Hilhorst,Henk W.M..
The remarkably short storability of recalcitrant seeds has prevented their inclusion in programs of ex situ conservation. The causes of their desiccation sensitivity and rapid decline in viability during storage are not fully elucidated yet. In this study the highly recalcitrant, fully viable embryos of the tree species Inga vera subsp. affinis were stored under various conditions and analyzed physiologically and cytologically at intervals, in order to obtain more insights in their loss of viability during storage. Embryos stored fully hydrated at 5ºC fully lost viability in 18 d. Sealed storage of partially dehydrated embryos slightly prolonged their viability, with 95% germination being attained after 14 d of storage. However, after 30 d of storage,...

Brighenti,Alberto Fontanella; Würz,Douglas André; Pasa,Mateus da Silveira; Rufato,Leo.
Abstract: The objective of this work was to investigate the effect of plant growth regulators for enhancing fruit color of 'Gala Standard' apples (Malus domestica). The experiment was carried out in the 2015 and 2016 crop seasons. The treatments consisted of water, as a control; 300 mg L-1 ethephon, as a positive control; 400 mg L-1 prohydrojasmonate; and 400 mg L-1 abscisic acid. Flesh firmness, soluble solids content, fruit weight, and red color were assessed after harvest. Plant growth regulators enhanced red color of fruit and chlorophyll degradation. Prohydrojasmonate and abscisic acid did not reduce flesh firmness, in the 2016 season.

Abstract: Somatic embryogenesis from explants from hermaphrodite papaya mother plants is an alternative for the production of true-to-type plants without the need for sexing. This study aimed to analyze hormonal and osmotic inducers in different somatic embryogenesis stages in the commercial hermaphrodite hybrid papaya UENF/Caliman 01. Leaf disks from in vitro shoots originated from ex vitro hermaphrodite plants were cultured in induction medium supplemented with different concentrations of 2,4-D (6, 9, 12, 15, and 18 μM) and 4-CPA (19, 22, 25, 28, and 31 μM). After 90 days, the formation of somatic embryos was verified. The 2,4-D induced the formation of light brown calli with low frequency (20%) of somatic embryogenesis. However, 4-CPA (25 μM) induced...

Variation in phytate accumulation in common bean (Phaseolus vulgaris L.) fruit explants BABT
Coelho,Cileide Maria Medeiros; Bellato,Cláudia Mattos; Garcia,Andréia Karime Marcelino; Vitorello,Victor Alexandre; Azevedo,Ricardo Antunes.
The in vitro synthesis of phytate was studied in common bean fruit explants. Different concentrations of sucrose; phosphorus (P); myo-inositol; abscisic acid (ABA); glutamine and methionine, were tested. Fixed concentrations of these compounds were tested at different periods (0, 3, 6 and 9 days). Variation in phytate coincided with different concentrations of sucrose, myo-inositol, P and ABA for the duration tested. These compounds caused an accumulation of phytate and were more effective in the presence of myo-inositol and P. The accumulation of P varied less than phytate for the different treatments tested in vitro. In conclusion, P, sucrose, ABA, and myo-inositol caused an increase in the phytate of bean seed, showing that it could be possible to alter...

Abstract Auxin regulates a plethora of events during plant growth and development, acting in concert with other phytohormones. YUCCA genes encode flavin monooxygenases that function in tryptophan-dependent auxin biosynthesis. To understand the contribution of the YUCCA4 (YUC4) gene on auxin homeostasis, plant growth and interaction with abscisic acid (ABA) signaling, 35S::YUC4 seedlings were generated, which showed elongated hypocotyls with hyponastic leaves and changes in root system architecture that correlate with enhanced auxin responsive gene expression. Differential expression of PIN1, 2, 3 and 7 auxin transporters was detected in roots of YUC4 overexpressing seedlings compared to the wild-type: PIN1 was down-regulated whereas PIN2, PIN3 and PIN7...
